The excitatory effect of glutamate and aspartate on central nervous system neurons described in the early 1960s (Curtis and Watkins, 1960; Krnjevic and Phillis, 1963) initiated a massive research effort aimed at proving these compounds as important neurotransmitters. During the 1980s, glutamate gained the status of the major excitatory neurotransmitter in the brain. Receptors were classified as N-methyl-Daspartate (NMDA) and non-NMDA receptors, based on electrophysiological studies (Hicks et al., 1978; Davies et al., 1979; Watkins and Evans, 1981; McLennan and Liu, 1982). NMDA receptors are stimulated most potently by NMDA itself. Currents activated by NMDA receptors are carried by Na § K § and Ca 2§ (Dingledine, 1983; MacDermott et al., 1986). The non-NMDA receptors were later divided into three groups, t~- amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors, kainate receptors, and metabotropic glutamate receptors, based on the preferred agonists, AMPA, kainate, and glutamate (for a review, see Watkins et al., 1990). AMPA and kainate receptors are ion channel coupled mainly Na § channels, whereas metabotropic glutamate receptors mediate responses via G-protein-coupled second messengers.

Research on, and interest in, red blood cell formation spans several centuries and was thought to have peaked in the 1980s with the cloning of the erythropoietin (EPO) gene. In the years subsequent to the cloning of EPO and its expression as a recombinant protein, much was written about EPO. Although much has been learned and published, new, exciting data are becoming available on almost a daily basis. Erythropoietins and Erythropoiesis: Molecular, Cellular, Preclinical, and Clinical Biology compiles both pertinent historical and very recent research on this molecule and its clinical utility. The book is divided into two sections: Background and Basic Science and Clinical Uses of Recombinant Erythropoietins. To begin, Israels and Israels describe the biology of red cells, the hierarchy of erythropoietic progenitor cells, their development to mature cells, and the effects of endogenous EPO on their development. Foote summarizes the historical interest in, and search for, an erythropoietic factor. Once EPO was identified, cloned, and expressed, the path was set for the study of other aspects of EPO biology both within erythropoiesis and other cellular systems.

Aging has long since been ascribed to the gradual accumulation of DNA mutations in the genome of somatic cells. However, it is only recently that the necessary sophisticated technology has been developed to begin testing this theory and its consequences. Endotoxins are potentially toxic compounds produced by Gram-negative bacteria including some pathogens. Unlike exotoxins, which are secreted in soluble form by live bacteria, endotoxins are comprised of structural components of bacteria. Endotoxins can cause a whole-body inflammatory state, sepsis, leading to low blood pressure, multiple organ dysfunction syndrome and death. This book brings together contributions from researchers in the forefront of these subjects. It is divided into two sections. The first deals with how endotoxins are synthesized and end up on the bacterial surface. The second discussed how endotoxins activate TLR4 and, in turn, how TLR4 generates the molecular signals leading to infectious and inflammatory diseases. In this book, seven chapters describe studies aimed at understanding and exploiting the key features of such molecular RNA and DNA devices. In the first section of the book, four chapters are devoted to artificial nucleic acid switches and sensors. These chapters introduce the concept of allosteric ribozymes as molecular switches and sensors; describe nucleic acid enzymes that are switched by oligonucleotides and other nucleic acid enzymes that are switched by proteins; and illustrate how switching elements can be integrated ration-ally into fluorescently signaling molecular sensors made out of nucleic acids. In the second section of the book, three chapters show that nature has been as crafty a molecular-scale engineer as any modern scientist via evolution of natural nucleic acid switches and sensors. RNAs have been found whose activities are modulated either by proteins or by small-molecule metabolites, and both kinds of system are described. Finally, the notion of exploiting naturally occurring RNA switches for drug development is discussed.
